CJC-1295 without DAC / Ipamorelin · Modified GRF (1-29) + Ipamorelin
Combination of a GHRH analog (without DAC) + selective GH secretagogue (GHS-R1a)
This presentation combines two peptides that act on complementary pathways of growth hormone release.
CJC-1295 without DAC, also called modified GRF (1-29), is the tetrasubstituted form of GHRH (1-29): D-alanine at position 2, glutamine at 8, alanine at 15 and leucine at 27. It lacks the DAC group that, in the original CJC-1295, lets it bind to albumin and extends its presence in plasma. It acts on the GHRH receptor of the pituitary. Ipamorelin is a pentapeptide agonist of the ghrelin receptor (GHS-R1a), described as the first selective GH secretagogue. There are no published studies of this blend; the evidence comes from each component and from studies combining GHRH with GH-releasing peptides.

Summary of results reported in the scientific literature. Each finding describes what was observed in the cited study, in its specific population and conditions; it does not constitute a therapeutic indication.
In its original characterization in rats, CJC-1295 was described as a tetrasubstituted form of hGRF (1-29); the DAC version, which binds to albumin, remained in plasma for more than 72 h. The CJC-1295 without DAC in this blend keeps the tetrasubstituted sequence without that group.
Jetté L et al. Endocrinology. 2005;146:3052-3058 · PMID 15817669In 10 healthy older men, 14 days of GHRH (1-29), the sequence CJC-1295 is derived from, given subcutaneously twice daily at the high dose, raised 24-hour GH and IGF-1 to values not significantly different from those of young men.
Corpas E et al. J Clin Endocrinol Metab. 1992;75:530-535 · PMID 1379256In rat pituitary cells, in rats and in pigs, ipamorelin has shown GH release with potency and efficacy comparable to GHRP-6. In pigs, it did not significantly raise ACTH or cortisol relative to GHRH, even at doses more than 200 times the ED50 for GH.
Raun K et al. Eur J Endocrinol. 1998;139:552-561 · PMID 9849822In healthy volunteers, ipamorelin has shown dose-proportional pharmacokinetics, with a terminal half-life of about 2 hours and a single episode of GH release per administration.
Gobburu JV et al. Pharm Res. 1999;16:1412-1416 · PMID 10496658In 18 healthy men, combining GHRH with submaximal doses of a GH-releasing peptide (GHRP-6, which acts on the same receptor as ipamorelin) has shown synergistic GH release.
